Open up MakeCD. Add all the files with Add->FS->Add and give it the directory in RAM. If you know your way around MakeCD all good.
From same ISO image prefs window check that Amiga settings and Rock Ridge are set from ISO button window. From Boot button window, enable MSDOS booting, image is OS4 CD L/slb_v2 file and emulation should be none. ...
Back to this business again! Where do I get that 'slb_v2' file? It is not on the original OS4.1 CD image I downloaded, at least in the System/L drawer... (there is no 'L' directory at the root level of 4.1 installation CD).

What machine are you playing with? Creating a whole new cd just to update the kickstart files might be unnecessarily. And the slb thing is not for all ng machines so not included with some systems. (i dunno what makes them bootable tough)
On my X5000 all i need is to insert some usb stick with a directory called "Kickstart" and power on and then load the updated kickstart files from there and continue to boot from any other media. Or i could just burn them to a cd-r without any additional settings needed afaik but i have not used that methood myself.
